Abstract
Cilj istraživanja: Cilj ovog istraživanja bio je utvrditi antitumorske učinke tretmana matičnih stanica karcinoma dojke novosintetiziranim inhibitorom fosfolipaze, spojem tieno [2,3-b] piridinom, preciznije, njegovo djelovanje na metastatski biljeg CD15s na MDA-MB-231 staničnoj liniji raka dojke. Materijali i metode: Trostruko negativna stanična linija raka dojke MDA-MB-231 tretirana je novosintetiziranim tieno-piridinskim spojem (3-amino-5-okso-N-naftil-5,6,7,8-tetrahidrotieno [2,3-b] kinolin-2-karboksamid) koncentracije 2 µM, tijekom 48h, kako bi se utvrdio postotak subpopulacije matičnih stanica raka pozitivnih na biljeg CD15s i na prosječni izražaj CD15s biljega po jednoj matičnoj stanici raka nakon tretmana. Nakon imunobojenja, pozitivne stanice detektirane su metodom protočne citometrije. Rezultati: Tretman novosintetiziranim inhibitorom PLC značajno je smanjio postotak subpopulacije matičnih stanica pozitivnih na biljeg CD15s, ali nije imao utjecaja na prosječni izražaj CD15s biljega po jednoj matičnoj stanici. Zaključci: Zbog utvrđenog smanjenja postotka subpopulacije matičnih stanica trostruko negativnog raka dojke koje na svojoj površini eksprimiraju metastatski biljeg CD15s, novosintetizirani inhibitor fosfolipaze C iz skupine tieno [2,3-b] piridina potencijalni je lijek za trostruko negativni karcinom dojke.
Abstract (english)
The aim of this study: The aim of this study was to determine the effects of treating of breast cancer stem cells with the newly synthesized thieno[2,3-b]pyridine anticancer agent, more precisely, focusing on its effect on metastatic marker, CD15 glyconjugate, expressed on the surface of cacer stem cells. Methods: The MDA-MB-231 triple-negative breast cancer cell line was treated during 48h with a 2 µM newly developed thienopyridine anticancer compound (3-amino-5-oxo-N-naphtyl-5,6,7,8-tetrahydrothieno[2,3-b]quinoline-2-carboxamide) to determine its effect on percentage of subpopulation of breast cancer stem cells that express CD15s and on the mean expression of CD15s on an individual breast cancer stem cells. Flow cytometric analysis was performed after immunostaining. Results: PLC inhibitor has significally decreased the percent of breast cancer stem cells positive on CD15s, but it was without effect upon mean expression of CD15s on an individual breast cancer stem cell. Conclusion: Due to its decreasing effect upon the percentage of triple negative breast cancer stem cells that are positive for the metastatic CD15s glycoconjugate, the investigated PLC inhibitor can be a potential drug for triple-negative breast cancer treatment.
