Abstract
CILJ ISTRAŽIVANJA: Cilj istraživanja je otkriti postoji li značajna razlika u izražaju autofagijskih gena p62, LC3 – A I LC3 - B u tumorskom tkivu mokraćnoga mjehura u odnosu na zdravo tkivo. USTROJ ISTRAŽIVANJA: Presječna studija. MJESTO ISTRAŽIVANJA: Istraživanje je provedeno u laboratoriju Katedre za imunologiju i genetiku, Medicinskog fakulteta u Splitu i na Odjelu za urologiju, Kliničkog bolničkog centra Split. SUDIONICI: Istraživanjem je obuhvaćeno 24 uzorka tkiva mokraćnoga mjehura. Od toga je bilo 12 uzoraka zdravog tkiva mjehura i 12 uzoraka adenokarcinoma mjehura niskog gradusa I stadija Ta. Svi uzorci uzeti su operacijskim zahvatom na Odjelu za urologiju, KBC-a Split. GLAVNA MJERA ISHODA: Glavna mjera ishoda je povećani izražaj autofagijskih gena u tkivu adenokarcinoma mokraćnoga mjehura, dokazana reakcijom RT-PCR. REZULTATI: Istraživanjem je obuhvaćeno 24 uzorka tkiva. Od toga je bilo 12 tumorskog tkiva, a 12 zdravog tkiva mokraćnoga mjehura. Obradom podataka, nismo dobili statistički značajan porast LC3 – A, LC3 - B ni p62 u tumorskom tkivu mokraćnoga mjehura (P<0,05). Dobili prosječni porast izražaja gena LC3-A i p62 ZAKLJUČCI: Ne postoji statistički značajan porast LC3 – A, LC3 – B ni p62 u tumorskom tkivu mokraćnoga mjehura ali s obzirom na povećanje prosječnog izražaja gena LC3-A i p62 možemo povezati autofagiju s nastankom karcinoma mokraćnoga mjehura.
Abstract (english)
OBJECTIVE: The aim of the study is to determine expression level of autophagic genes p62, LC3 – A and LC3 - B in bladder carcinoma (compared to the healthy bladder tissue). DESIGN: The study was designed as a cross-sectional study. SETTINGS: The study was carried out at the Department of Imunology and Genetics, at the Medical University Split and the Department of Urology, University Hospital Centre Split. PARTICIPANTS: The study consisted 12 was bladder carcinoma tissue, and the 12 healthy bladder tissue samples. All of the samples were collected during an operational procedure at the Department of Urology, University Hospital Centre Split. MAIN OUTCOME MEASURES: Main outcome measure is a elevated expression of autophagic genes in the bladder carcinoma tissue, which has been detected by RT-PCR approach. RESULTS: We have not found a statistically significant increase of LC3 – A, LC3 – B or p62 expression in the tumor tissue, compared to the healthy bladder tissue (P<0,05). The increase in genes expression was not statistically significant. We got average increase of gene expression in LC3-A and p62. CONCLUSIONS: We have not found a significant increase in LC3 – A, LC3 – B or p62 in tumor tissue but average increase of gene expression in LC3-A and p62 indicates connection between autophagy and bladder carcinogenesis.
